Longtemps consid\u00e9r\u00e9e comme une pathologie purement neurologique, cette maladie pourrait en r\u00e9alit\u00e9 d\u00e9buter\u2026 dans le ventre.<\/p>\n\n\n\n<p>Une \u00e9tude r\u00e9cente publi\u00e9e dans <em>Nature Communications<\/em> (novembre 2023), relay\u00e9e par <em>Sciences et Avenir<\/em>, vient confirmer ce lien, en montrant que les personnes atteintes de Parkinson pr\u00e9sentent un <strong>d\u00e9s\u00e9quilibre important du microbiote intestinal<\/strong>. D\u00e9cryptage.<\/p>\n\n\n\n<hr class=\"wp-block-separator has-alpha-channel-opacity\"\/>\n\n\n\n<h2 class=\"wp-block-heading\">\ud83d\udd0d Une intuition ancienne qui se confirme<\/h2>\n\n\n\n<p>En 2003, le neurologue allemand Heiko Braak formule une hypoth\u00e8se encore jug\u00e9e audacieuse \u00e0 l\u2019\u00e9poque : <strong>la maladie de Parkinson pourrait na\u00eetre dans le syst\u00e8me digestif<\/strong>, avant de progresser vers le cerveau via le nerf vague \u2014 une sorte d\u2019autoroute neuronale reliant les intestins au tronc c\u00e9r\u00e9bral.<\/p>\n\n\n\n<p>Depuis, les \u00e9tudes se multiplient et convergent :<\/p>\n\n\n\n<ul class=\"wp-block-list\">\n<li>En 2017, des chercheurs su\u00e9dois montrent que <strong>les personnes ayant subi une vagotomie<\/strong> (section du nerf vague) ont <strong>moins de risques de d\u00e9velopper Parkinson<\/strong>.<\/li>\n\n\n\n<li>Les troubles digestifs chroniques (constipation, ballonnements, inflammation) <strong>pr\u00e9c\u00e8dent souvent de plusieurs ann\u00e9es les sympt\u00f4mes moteurs<\/strong> (tremblements, raideur).<\/li>\n<\/ul>\n\n\n\n<hr class=\"wp-block-separator has-alpha-channel-opacity\"\/>\n\n\n\n<h2 class=\"wp-block-heading\">\ud83e\udda0 Un microbiote profond\u00e9ment d\u00e9s\u00e9quilibr\u00e9<\/h2>\n\n\n\n<p>Dans la derni\u00e8re \u00e9tude de l\u2019Universit\u00e9 de l\u2019Alabama, les chercheurs ont compar\u00e9 les microbiotes intestinaux de <strong>490 patients parkinsoniens<\/strong> et de <strong>230 t\u00e9moins sains<\/strong>, dont beaucoup \u00e9taient les conjoints des malades.<\/p>\n\n\n\n<p>Les r\u00e9sultats sont sans appel :<\/p>\n\n\n\n<ul class=\"wp-block-list\">\n<li>Une <strong>surabondance de bact\u00e9ries pathog\u00e8nes opportunistes<\/strong> ;<\/li>\n\n\n\n<li>Une <strong>diminution des bact\u00e9ries b\u00e9n\u00e9fiques<\/strong>, notamment celles impliqu\u00e9es dans la <strong>r\u00e9gulation de l\u2019inflammation<\/strong> ;<\/li>\n\n\n\n<li>Un microbiote globalement <strong>plus inflammatoire et d\u00e9s\u00e9quilibr\u00e9<\/strong> chez les patients.<\/li>\n<\/ul>\n\n\n\n<p>Cette composition intestinale pourrait <strong>favoriser un climat inflammatoire chronique<\/strong>, propice \u00e0 des d\u00e9sordres neurologiques.<\/p>\n\n\n\n<hr class=\"wp-block-separator has-alpha-channel-opacity\"\/>\n\n\n\n<h2 class=\"wp-block-heading\">\ud83d\udd2c Une piste biologique : la prot\u00e9ine \u03b1-synucl\u00e9ine<\/h2>\n\n\n\n<p>Un autre marqueur cl\u00e9 de Parkinson est la <strong>prot\u00e9ine \u03b1-synucl\u00e9ine<\/strong>, qui s\u2019agglutine de fa\u00e7on anormale dans les neurones des patients.<\/p>\n\n\n\n<p>Or, des \u00e9tudes montrent que cette prot\u00e9ine <strong>peut s\u2019accumuler dans l\u2019intestin<\/strong> avant d\u2019atteindre le cerveau. Pire : certaines bact\u00e9ries, comme <em>E. coli<\/em>, produisent des <strong>prot\u00e9ines amylo\u00efdes bact\u00e9riennes<\/strong>, qui <strong>favorisent l\u2019agr\u00e9gation pathologique de l\u2019\u03b1-synucl\u00e9ine<\/strong>. Ce ph\u00e9nom\u00e8ne pourrait initier la cascade neurod\u00e9g\u00e9n\u00e9rative.<\/p>\n\n\n\n<hr class=\"wp-block-separator has-alpha-channel-opacity\"\/>\n\n\n\n<h2 class=\"wp-block-heading\">\ud83d\udc8a Le microbiote influe aussi sur les traitements<\/h2>\n\n\n\n<p>La <strong>L-Dopa<\/strong>, mol\u00e9cule phare du traitement de Parkinson, est cens\u00e9e atteindre le cerveau via la circulation sanguine. Mais l\u00e0 encore, le microbiote peut interf\u00e9rer.
